Eva Mendes è nata a Miami, Florida, nel 1974 (anche se alcune fonti citano erroneamente Houston, 1978  ) in una famiglia cubana, e ben presto si trasferì con sua madre (dopo il divorzio della stessa) a Los Angeles. Suo padre è un venditore d'auto e sua madre, anch'essa di nome Eva, è la preside di una scuola elementare.

Eva iniziò la sua carriera dai video commerciali ai videoclip musicali fino alle apparizioni nelle soap opera e nei telefilm, e qualche piccolo ruolo secondario. Ha ricevuto il suo primo vero ingaggio nell'acclamato Training Day, dove compare completamente nuda. Seguirono in breve tempo Fratelli per la pelle, Once Upon a Time in Mexico e Hitch, dove ebbe la parte femminile accanto a Will Smith. Once Upon a Time in Mexico le ha permesso di ricevere una nomination ai Teen Choice Awards.

Nel 2006 la famosa rivista Maxim, tra le Hot 100, l'ha segnalata al numero 27.

Mendes è legata sentimentalmente al regista peruviano George Gargurevich. Vive nelle Hollywood Hills. I suoi film preferiti sono Shining (1980) e Blow-Up (1966).
