Nata in California, si trasferì a Tucson (Arizona). Il padre era irlandese (è morto per infarto quando Kate aveva 22 anni) e la madre è italiana.

Ha iniziato la sua carriera da attrice nel teatro dell'Universitá dell'Arizona per poi proseguire a Chicago, dove ha studiato alla rinomata Piven Theatre Workshop. A Chicago ha recitato in due film indipendenti, Normal Life diretto da John McNaughton, e Henry, Portrait Of A Serial Killer II. La sua carriera ha decollato rapidamente in seguito a un trasferimento a New York per formarsi in commedia e improvvisazione, dove ottiene una serie di ruoli regolari in "The Mike O'Malley Show", "Mind of a Married Man" e "The Drew Carey Show". Nei Primi anni '80 é stata una nota modella in Giappone, dove era venerata per la sua notevole statura: 1,78 m.

In Italia e nella Svizzera italiana è famosa soprattutto per l'interpretazione di Addison Forbes Montgomery, chirurgo neonatale, nella serie televisiva trasmessa da Italia Uno e dalla rete satellitare SKY, Grey's Anatomy. Ha partecipato anche ad altri telefilm quali CSI: Crime Scene Investigation e Law & Order. Attualmente è impegnata nelle riprese della serie Private Practice, spin-off di Grey's Anatomy.
